Cell Type-Specific Promoters of Volvox carteri for Molecular Cell Biology Studies

چکیده

The multicellular green alga Volvox carteri has emerged as a valuable model organism for investigating various aspects of multicellularity and cellular differentiation, photoreception phototaxis, cell division, biogenesis the extracellular matrix morphogenetic movements. While range molecular tools bioinformatics resources have been made available exploring these topics, establishment type-specific promoters in V. not achieved so far. Therefore, here, we conducted thorough screening transcriptome data from RNA sequencing analyses order to identify potential promoters. Eventually, chose two putative strong promoters, with one exhibiting specific expression reproductive cells (gonidia), PCY1 promoter, other somatic cells, PFP promoter. After cloning both promoter regions, they were introduced upstream luciferase reporter gene. By using particle bombardment, DNA constructs stably integrated into genome carteri. results analyses, which at transcript protein levels, demonstrated that drive their respective target types. Transformants considerably diverse levels chimeric genes identifiable. In conclusion, analysis allowed identification Reporter gene actual usability investigated proven be potent genetic engineering

Reporter genes and highly regulated promoters as tools for transformation experiments in Volvox carteri.

The multicellular alga Volvox is an attractive model for the study of developmental processes. With the recent report of successful transformation, regulated promoters as well as reporter genes working in this organism are now required. The Volvox genes encoding arylsulfatase and the extracellular glycoprotein ISG are strictly regulated. The former is transcribed only under conditions of sulfur...

The relationship between cell size and cell fate in Volvox carteri

In Volvox carteri development, visibly asymmetric cleavage divisions set apart large embryonic cells that will become asexual reproductive cells (gonidia) from smaller cells that will produce terminally differentiated somatic cells. Three mechanisms have been proposed to explain how asymmetric division leads to cell specification in Volvox: (a) by a direct effect of cell size (or a property der...

Sexual Induction in Volvox Carteri

The study of the dependence of reactions on the concentration of various compounds is often of great value in determining reaction mechanisms . In chemical kinetics, it is often found that, according to van't Hoff's rule, the order of dependence of a reaction on the concentration of some compound involved in that reaction is equal to the number of molecules of the compound involved in each reac...

A revision of the cell lineages recently reported for Volvox carteri embryos

Volvox offers a unique opportunity to study the segregation of reproductive potential during development of a multicellular organism. At the sixth cleavage division of an asexual embryo of Volvox carteri, strain HK 10, one half of the 32 cells typically cleave unequally to yield 16 large gonidial (reproductive) initials and 16 smaller somatic initials, while the remaining cells divide equally t...

A Preliminary Genetic Investigation of VOLVOX CARTERI.

A preliminary genetic analysis of a number of genetic variants of Volvox carteri f. nagariensis is presented. Techniques are outlined for mutagenesis of Volvox, isolation of mutants and routine genetic analysis. All of the mutants show simple Mendelian segregation patterns and have been tentatively placed in 14 linkage groups.
